Περίληψη: Organisms have “safety valves” to maintain the integrity of their proteome. The protein homeostasis (proteostasis) network (PN) works accurately to facilitate the proper balance between protein synthesis and clearance. There is a gradual loss of protein quality control during aging progression due to the impairment of various proteostatic mechanisms. Disruption of proteostasis leads to aberrant accumulation of misfolded proteins that are targets of the cellular degradation machineries; the ubiquitin–proteasome system (UPS) and the autophagy-lysosomal pathway (ALP). Due to the age-related dysfunction of both UPS and ALP, misfolded proteins remain undegradable and form toxic aggregates that further inhibit the proteasome and negatively affect autophagy. This vicious cycle further aggravates the manifestation of various age-related protein-misfolding diseases (PMDs). This chapter shortly discusses the protective mechanisms of protein quality control and mainly focuses on the UPS. Since UPS impairment is associated with PMD progression, we refer to the UPS status in aging and age-related diseases and its implication in selected PMD pathology. We also discuss the interventions that have been performed toward proteasome activation as a promising therapeutic approach for the treatment of the major PMDs, such as various neurodegenerative diseases.
