Περίληψη: | The AtoS-AtoC system in E. coli is a member of the two-component system (TCS) family. Upon induction by external stimuli, the membrane sensor kinase AtoS autophosphorylates and subsequently it phosphorylates the response regulator AtoC, which activates the transcription of the atoDAEB operon encoding for proteins involved in short-chain fatty acid catabolism. Additionally, the AtoS-AtoC system and its target atoDAEB operon regulate the complexed poly-(R)-3- hydroxybutyrate (cPHB) biosynthesis under acetoacetate or spermidine induction. Global analyses of the E. coli TCSs have revealed that AtoS-AtoC might be involved in many additional processes. This study aimed to investigate the possible contribution of the AtoS-AtoC TCS in E. coli chemotaxis. E. coli DatoSC and atoSC+ and their transformed derivatives with plasmids overproducing the whole or part of the AtoS-AtoC TCS, were grown with Asp, Ser, Met or Glu as chemoattractants, in the presence of acetoacetate, spermidine, histamine, C48/80 or Ca2+. Chemotaxis was evaluated by determining cell motility using the swarm agar assay. DatoSC cells were not motile independently to the chemoattractant or the agent added. The extrachromosomal introduction of the AtoS-AtoC system in DatoSC bacteria restored their motility. The AtoS-AtoC expressing cells demonstrated an agent concentration-dependent motility in the presence of Asp and Ser. DatoSC cells expressing either AtoS or AtoC in trans demonstrated no response to the chemoattractants independently of the agents added. These data suggested that the AtoS-AtoC TCS may participate in the E. coli chemotactic response, thus being implicated in bacterial adaptation to continuously changing environments. |
